Assay Principle
AMP Deaminase (AMPD) catalyzes the hydrolytic deamination of adenosine monophosphate (AMP) to inosine monophosphate (IMP) and ammonia. The PRECICE® AMP Deaminase Assay Kit measures this activity through a coupled enzymatic reaction:
- 1. AMP Deaminase Reaction:
AMP + H₂O → IMP + NH₄⁺ - 2. IMP Dehydrogenase Reaction:
IMP + NAD⁺ → XMP + NADH + H⁺
The formation of NADH is continuously monitored by the increase in absorbance at 340 nm, providing a direct and quantitative measure of AMP deaminase activity.
