IMPDH Inhibition
Aim: Screening of compounds for their abilities to inhibit Human Inosine Monophosphate Dehydrogenase II in vitro. Determination of the inhibition kinetics of a given compound and measurement of its IC50 value.
IMPDH enzyme: The enzyme used in the assays is a Human recombinant Inosine Monophosphate Dehydrogenase (IMPDH, Type 2, Ref. E-Nov1), produced and purified by NOVOCIB. Positive control consists in inhbiting IMPDH II with a 8- or 12-concentration range of mycophenolic acid (MPA).
IMPDH in vitro screening can also be performed on a bacterial recombinant IMPDH from Staphylococcus aureus, expressed in E. coli (produced and purified by NOVOCIB, Ref. E-Nov7).

Positive control consists in inhbiting PNP with a 8- or 12-concentration range of Ganciclovir. Other validated PNP inhibitor can be used as positive control when provided.

AK enzyme: The enzyme used in the assays is a Human recombinant Adenosine Kinase (Ref. E-Nov5), produced and purified by NOVOCIB. Positive control consists in inhbiting AK with a 8- or 12-concentration range of A-134974 (Sigma-Aldrich, Ref. A2846).

Nucleoside Kinase - IMPDH Coupled Assay (^Top)
Aim: Rapid evaluation of monophosphate forms of nucleoside analogues as IMPDH inhibitors. In vitro Phosphorylation of nucleoside analogues by Adenosine Kinase or cytosolic 5'-nucleotidase and screening of monophosphorylated forms for their abilities to inhibit Human IMPDH.
Enzymes: Adenosine Kinase (AK, Ref. E-Nov5), or cytosolic 5'-nucleotidase II (cN-II, Ref. E-Nov6) and Inosine Monophosphate Dehydrogenase, Type II (Ref. E-Nov1) are Human recombinant enzymes, produced and purified by NOVOCIB. Positive control consists in inhbiting IMPDH II with ribavirin-MP obtained from ribavirin phosphorylation by AK.
